With a lens, sharpness does not decrease as exposure time increases. Unless
you are referring to the loss of sharpness from subject movement or loss of
resolution on the film from overexposure...

Smaller holes do (not may) lead to decreased sharpness because of
diffraction, and yes it is visible depending on the enlargement.
