You just use the clean plexi like you would use a final transfer paper:
Transfer the tissue in cold water, squeegee, let sit for 5-10 minutes,
develop. If you find the tissue hard to peel off during development, try a
1-2 min. presoak in rubbing alcohol before development. The gelatin will
stick to almost any smooth, waterproof surface, but plexi will release the
relief again.
But you should first try to master the single transfer method before
switching to double transfer...

Not at all. But it must be cleaned well. Some say, it should be slightly
sanded down, some think the glossy surface does as well. It probably
doesn't matter... When I used fresh plexi, it worked great, after some use
I got some problems, then sanded it down, worked fine for a while, then
again I got occasional problems. Sounds like some contamination, although I
was really rubbing like crazy...
